MDM_2癌基因cDNA探针制备及在肿瘤组织中扩增的检测

PREPARATION OF MDM_2 ON COGENE cDNA PROBE AND ITS DETECTION IN TUMOR TISSUES

为了研究ＭＤＭ＿２癌基因在人自发肿瘤中的分子改变，用逆转录ＰＣＲ法扩增出人ＭＤＭ＿２ＤＮＡ片段，将其克隆于质粒ｐＵＣ＿１８，完成了探针的制备，并检测了２０例人间叶肿瘤，发现其中７例肉瘤有不同程度ＭＤＭ＿２扩增，而３例良性者则为阴性。ＭＤＭ＿２基因改变与人肉瘤发生的关系尚待进一步研究。

To further detect the changes in MDM_2 gene of human spontaneous sarcomas,human MDM_2 cDNA was amplified by RT-PCR and subcloned it in plasmid pUC_18.Usingthe probe ,we checked 20 cases of human mesenchymal tumors.of which,7 sarcomasshowed amplification with different extent , while all of 3 benign tumors gave negetive re-sults.The relationship between MDM_2 gene alteration and progression of human sarcomasshould be further studied.
