卡铂对A_(549)肺癌细胞的抗癌作用及其机制的初步探讨

摘要 本文观察了卡铂对A_(549)肺癌细胞DNA合成、DNA链间交联的影响及细胞超微结构的改变。依卡铂剂量不同将实验分为A、B、C、D四组,剂量分别为20μg/ml、40μg/ml、60μg/ml、80μg/ml,另设空白对照E组。结果表明,卡铂可能是DNA模板损伤型药物,通过引起DNA交联而阻碍其复制,产生杀细胞效应。 In order to study the anticancer effects of carboplatin (CBDCA) in vitro.We have examined the inhibition of DNA synthesis,DNA interstrand cross-linking(ISC)and the changes of ultrastructure of A549 Lung cancer cells (A549 cells).Four experimental groups, named A (20ug/ml)B(40ug/ ml),C(60ug/ml) and D (80ug/ml)according to different dosage of CBDCA and one group without CBDCA as control were used. In conclusion,we demonstrate that CBDCA is a DNA breakage analog,The DNA ISC induced by CBDCA is related to the inhibition of DNA replication and cytotoxicity.
