反相高效液相色谱法测定洛美沙星的血和尿药浓度

摘要 本文报道用反相高效液相色谱法测定血清和尿液中洛美沙星浓度,固定相用C_(18)柱,流动相为甲醇/0.008M磷酸盐缓冲液/0.5M四丁基溴化铵(25:75:4,V/V/V).荧光检测激发波长280nm,发射波长455nm,以外标法定量。洛美沙星的最低检出限为0.06μg/ml,线性范围为0.125-8.μg/ml.线性关系良好(r=0.9999),日内变异系数2.87%,日间变异系数2.94%,回收率75.8%~80.9%。 Reversed-phase HPLC was described for the separation and determination of Iomefloxacin inserum and urine. Samples were analysed on an ODS column (7μ, 250×4.6mm) with a mobile phase ofmethand-0.008M phosphate buffer-0.5M tetrabuty lammonium bromide(25: 75 :4,v/v/v).usingfluorescence detection at λ ex 280nm and λ em 455nm.The detection limit was 0. 06μg/ml. GoodIinearity was obtained in the concentration range from 0. 125 to 8μg/ml with the correlation coeffi-cient r= 0. 9999. The average recovery was 75.8%-80.9%and the differences within and betweendays were 2. 87%,2. 94% respectively
