强力宁对慢性活动性肝炎患者IL-6、TNF-α影响的体外研究

摘要 为了解甘草甜素对慢性活动性肝炎(CAH)患者外周血单个核细胞(PBMC)产生的肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)的影响,用强力宁(主要成分为甘草甜素)对18例CAH患者PBMC产生IL-6、TNF-α的能力进行了研究。结果显示在体外培养条件下,强力宁可使PBMC产生IL-6、TNF-α的能力减弱;而且PBMC产生IL-6、TNF-α的能力降至最低(接近正常)时所需的剂量在CAH活动期明显低于稳定期(P<0.01)。提示用强力宁治疗CAH时应根据不同病程调节剂量。研究尚发现强力宁可能具有诱生PBMC膜产生可溶性IL-6受体的作用,从而中和了培养上清液中的IL-6。上述结果可部分解释其治疗CAH的机理。 To investigate the effect of glycyrrhizin on peripheral blood mononuclear cells(PBMC)interleukin-6( IL-6) and tumor necrosis factor-α(TNF-α) levels of patientswith chronic active hepatitis(CAH),the effect of potenlin(the main composition ofglycyrrhizin)on the ability to produce IL-6 and TNF-α of PBMC in patients withCAH was studied.The result revealed that in vitro culture condition,potenlin maylead to a decrease of the ability of PBMC to produce TNF-α and IL-6. when the abi- lity of produeing IL-6 and TNF-αwas the lowest,the required minimum dose of po-tenlin active stage is lower than that in inactive stage .Above data suggested thatwhen CAH patients are treated with potenlin,the dose of potenlin varies with differentstages.It was also found that potenlin may induce PBMC membrane to produce solubleIL-6 receptors Which neutralize IL-6 in cultured supernatant,this may in part explainthe possible mechanism of potenlin in CAH treatment.
