氧氟沙星脂质体的制备及治疗兔实验性绿脓杆菌角结膜炎的研究

Preparation of Ofloxacin Liposomes and Treatment of Experimental Pseudomonas Kerato- conjunctivitis in Rabbits

氧氟沙星脂质体和无药脂质体由注入法制备，并用紫外分光光度计检测和光学显微镜观察。兔角膜实质接种绿脓杆菌，制成绿脓杆菌角结膜炎模型。无药脂质体为对照组，氧氟沙星脂质体和氧氟沙星滴眼剂为实验组。观察治疗前后结膜和角膜病变，并收集结膜囊分泌物进行培养。结膜病变显示，给药后第２、４、７天，两实验组与对照组相比都有显著性差异，第２天，氧氟沙星脂质体组明显好于氧氟沙星滴眼剂组。角膜病变显示，给药后第２、４、７天，氧氟沙星脂质体组好于对照组，有显著性差异。第７天，氧氟沙星滴眼剂组与对照组相比有显著性差异。细菌培养结果显示，给药后第２天，两实验组与对照组相比，有显著性差异。第７天，氧氟沙星脂质体组和氧氟沙星滴眼剂组的培养阳性率都增加了，但氧氟沙星滴眼剂组阳性率的增加程度高于氧氟沙星脂质体组。氧氟沙星脂质体和氢氟沙星滴眼剂都可用来治疗绿脓杆菌角结膜炎，氧氟沙星脂质体的作用优于氧氟沙星滴眼剂。

floxacin liposomes and empty liposomes were prepared by method of injection,then they were detectedby UV-visible spectrophotometer and observed by microscope. Pseudomonas aeruginosa was inoculated intocorneal stroma of rabbits and the model of keratoconjunctivitis was estabished.Empty liposomes were used ascontrol;ofloxacin liposomes and ofloxacin eyedrops groups were experimental ones.After six times's treat-ment,conjunctival and corneal lesions were examined,and swabs of conjunctival sac were cultured.Results:in conjunctiva,lesions of two experimental groups were all significantly less than those of control group onthe 2nd, 4th and 7th day after treatment.Lesions of ofloxacin liposomes group were also significantly lessthan those of ofloxacin eyedrops on 2nd day.In cornea,lesions of ofloxacin liposomes group were signif-cantly less than those of control group on 2nd,4th and 7th after treatment. Lesions of ofloxacin eyedropsgroup were significantly less than those of control group on 7th day. In result of bacterium culture,the cul-ture's positive rate of two experimental groups and control group had significant differences on 2nd day. On7th day,the positive rate of two experimental groups increased,but that of ofioxacin liposomes group hadless increase.It is concluded that ofloxacin liposomes or ofloxacin eyedrops had effects on treatment of pseu-domonas keratoconjunctivitis in the rabbit model,and the effect of ofloxacin liposomes was better than that ofofloxacin eyedrops.
