摘要
研究了不同膜(PS,PAN)的截流特性及pH、压强差、浓缩倍数对超氧化物歧化酶(SOD)分离纯化的影响.当采用PAN膜,浓缩4倍时,SOD收率为85%,比活提高到1350U/mg。本文还研究了热变性和超滤先后次序对SOD纯化的影响。结果表明先超滤、后热变性工艺,SOD纯化后总收率68%,比活达6075U/mg。
Purification of superoxide dismutase from Pig blood with hollow fiber ultrafiltration was studied in this paper. The membrane type, pH, concentration factor and heat treatment are discussed. When PAN membrane (molecular weight cut off 20000) was used to concentrate the supernatant of the red cell hemolysate for 4 times, thh SOD could be purified 12 times. The recovery and the specific activity of SOD after this process were 68% and 6075U/mg, respectively.
