粉防已碱对大鼠腹腔巨噬细胞释放部分炎症介质的影响 被引量:20

摘要 血小板活化因子(PAF)、肿瘤坏死因子(TNF)及白介素-1(IL-1)是几个很强的炎症介质,对多种细胞、组织及器官都有重要的作用。利用洗涤的兔血小板聚集的方法和酶联免疫法、小鼠胸腺细胞增殖法分别测定了粉防已碱(Tet)对脂多糖(LPS)刺激大鼠腹腔巨噬细胞(PMΦ)释放PAF、TNF及IL-1的影响,结果表明Tet对TNF、IL-1及PAF的释放都有显著地抑制作用,并在0.1~100μmol/L有剂量依赖关系,这提示Tet的抗炎、免疫抑制作用的机理之一是由于Tet抑制了PAF、IL-1、TNF等炎症介质的释放。 Platelet-activating factor (PAF),tumor necrosis factor(TNF)and interleukin 1(IL-1)are potent molecular mediators of imflammation and exert significant effects on various types of cells,tissues and organs.The effects of tetrandrine(Tet)on the PAF,TNF and IL-1 released by the peritoneal macrophages of rats were observed with the methods of washed rabbit platelets,mouse thymocyte proliferation and enzyme-linked immunosorbant assay respectively.It was found that the release of PAF,TNF and IL-1 were significantly inhibited by Tet in a dosage-dependent manner of the range 0.1~100 μmol/L,whch indicates that one of the mechanisms of the anti-inflammatory effect of Tet is its inhibition to the release of PAF,TNF and IL-1 by the peritoneal macrophages.
