摘要
为了探讨缺氧情况失血性休克的发生发展的机理,我们观察了在模拟海拔4000m高原失血性休克山羊的血浆(shockplasma,SP)对培养的肺动脉内皮细胞(pulmonaryarterialendothelialcell,PAEC)的作用。PAEC在含不同SP培养液中培养24h后进行观察,在高浓度SP条件下细胞收缩变圆、脱落,大部分细胞死亡,细胞活力明显降低,以上变化具有明显的浓度依赖关系(r=0.985,P<0.001)。加热灭活的SP或经鱼精蛋白沉淀的SP毒性作用明显减弱。PAEC在含50%SP培养液中培养1~3h后,细胞活力明显升高,3h达高峰,3~6h后明显降低,6h末降至最低值,12~24h末逐渐回升。上述结果表明,缺氧复合血性休克动物血浆中含有对PAEC激活和损伤作用的成份,其作用具有浓度依赖和时间依赖关系。该成份对热敏感,可被鱼精蛋白沉淀。本实验提示血管内皮细胞的“激活-损伤”可能参与缺氧复合失血性休克的发展过程。
The effects of plasma from the goats in hypoxia complicated with hemorrhagic shock on the cultured endothelial ces of pullmonary artery (PAEC)were observed.It was found that the goat pasma damaged,devitaized or decreased the viabiity of PAEC in a concentration-dependent parttern. The toxic effects of goat plasma was significantly decreased after heating (56 ℃ for 30 minutes)or precipitation with protamine.Goat pasma exerted diphasic time-dependent effects on PAEC,that is,the activation phase which lasted from the lst to 3rd hour after the administration of goat pasma and in whihc the viabiity of PAEC was increaeed and the damage phase which extended from the 3rd to 6th hour.These findings suggest that goat pasma during hypoxia compicated hemorrhagic shock may contain certain substances which can activate and damage PAEC and the activation phase of PAEC may pay an important roe in the pathoogica process of hemorrhagic shock.
