摘要
采用体外细胞培养、免疫细胞化学(ICC)染色和放射免疫(RIA)测定等技术,从定性和定量分析等方面对人脐静脉内皮细胞(HUVEC)及心钠素(ANP)进行研究,结果显示:体外培养第5天的HUVEC,ICC染色呈ANP样免疫阳性反应;RIA测定作为空白对照的未培养细胞的RPMI-1640细胞培养液,其中不含ANP,而培养了HUVEC的细胞培养上清液中ANP含量为25.2±2.9ng/10^5个细胞。
The endothelial cells of human fetal umbilical vein were cultured and immunocvtochemical staining showed positive atrial natriuretic polypeptide(ANP)-like immuno reaction after 5 days of culture. Meanwhile 25.2±2.9 ng of ANP/10 ̄5 endothelial cells of fetal urnbilical vein was found in the supernatant with radioimmunoassay but it was zero in the control.It is postulated that ANP in the supernatant of endothelial cell culture originates from the secretion of endothelial cells.
