体外培养人脐静脉内皮细胞合成与分泌心钠素的初步观察

Study of synthesis and secretion of atrial natriuretic polypeptide in the endothelial cells of human fetal umbilical vein in vitro

采用体外细胞培养、免疫细胞化学（ＩＣＣ）染色和放射免疫（ＲＩＡ）测定等技术，从定性和定量分析等方面对人脐静脉内皮细胞（ＨＵＶＥＣ）及心钠素（ＡＮＰ）进行研究，结果显示：体外培养第５天的ＨＵＶＥＣ，ＩＣＣ染色呈ＡＮＰ样免疫阳性反应；ＲＩＡ测定作为空白对照的未培养细胞的ＲＰＭＩ－１６４０细胞培养液，其中不含ＡＮＰ，而培养了ＨＵＶＥＣ的细胞培养上清液中ＡＮＰ含量为２５．２±２．９ｎｇ／１０＾５个细胞。

The endothelial cells of human fetal umbilical vein were cultured and immunocvtochemical staining showed positive atrial natriuretic polypeptide(ANP)-like immuno reaction after 5 days of culture. Meanwhile 25.2±2.9 ng of ANP/10￣5 endothelial cells of fetal urnbilical vein was found in the supernatant with radioimmunoassay but it was zero in the control.It is postulated that ANP in the supernatant of endothelial cell culture originates from the secretion of endothelial cells.
