摘要
作者采用自行设计的方法,首次在鼻咽癌病人血清中检测出凝集因子(AF)。鼻咽癌病人血清以D-Hanks缓冲液倍比稀释(1:20~2560)。人鼻咽癌细胞株(CNE-2)经胰蛋白酶消化,戊二醛固定制成单个细胞悬液。两者混合于96孔培养板,5000细胞/孔,经37℃、4℃、再37℃处理后,在倒置显微镜下观察,以能形成一个≥20细胞的凝集团块为AF阳性,结果发现231例鼻咽癌病人血清都是阳性,几何平均滴度(GMT)1:265.1。111名正常人中74人AF阴性,37人低滴度阳性(1:20~160),GMTI:39.2。提示鼻咽癌病人血清AF水平明显高于正常人(P<0.001)。作者对结果及方法进行了讨论。
Using a new method
established by themselves , the authors detected agglutination factor(AF)in sera of
nasopharyngeal carcinoma(NPC)patients.The sera of NPC patients were diluted by multiple
propor-tions(1:20~2560)with D-Hanks buffer solution. Single cell suspensions of CNE-2 cells
were prepared aftertrypsinization and glutaraldehyde fixation. Both of them were mixed up into
96 welI microculture pIate, 5000cells per well,then the plate was incubated at varying
temprature(37℃-4℃-37℃). The cell agglutinationwas observed under inverted microscope,so
long as formed one agglutinative cell clump(≥20 cells)in a well,the result was considered as
positive The results showed that in 23l cases of NPC AF were positive,geometri-cal mean
titre(GMT) was 1:265.1.Among 111 cases of healthy controls,74 were negative, 37 were
positivewith low titre(1:20~160), GMT was 1: 39.2.It is Obvious that the AF Ievels in serum of
NPC patientswere significant higher than that of the healthy controls(P<0.00.In this paper the AF
phenomenon andmethodology were discussed.
