摘要
本文以二种凝集素WGA、LCA为探针,通过SDS-PAGE、印迹转移及ABC免疫酶标染色方法,连续观察了着床期间(D5-D9,交配日为D0)兔子宫内膜糖蛋白表达的动态变化。发现WGA结合41Kd、85Kd,LCA结合45Kd、83Kd糖蛋白在胚泡进入子宫腔,但尚游离时(D5,D6)开始升高,在着床始发的D7天达高峰,胚泡植入后(D7.5,D8)下降,着床完成时(D9)多已消失。其中41KdWGA结合和45KdLCA结合特异性糖蛋白变化尤为突出,其含量在未孕时极微,D7显著增高,着床开始后(D7.5)即迅速下降,且这两条带主要存在于胚泡着床侧。结果表明:着床期间子宫内膜表面糖蛋白有阶段特异性变化,并该变化可能受到胚泡的局部调节作用。
BY SDS-PAGE,Western blot,and ABC immune enzyme-labled assay,we studied the dynamic change of glycoprotein expression on rabbit endometrium at peri-implantation stage.Lectins including WGA,LCA,were used as probes,The results showed that 41Kd and 85Kd WGA,45Kd and 83Kd LCA-binding glycoprbteins began to increase from preimplantation stage(D5,D6),and reached their highest levels at implantaton stage(D7),respectively,then decresed D7.5.D8,and disappeared at post-implantation(D9)stage.Among them,it was more noticeable that changes of two glycoproteins binding to WGA(41Kd)and to LCA(45Kd) were more obvious,and primarily seen on anti-mesometrial region.These reaults indicate that stage-specific change of glycoprotein expression occuring on rabbit endometrium, and may be related to the regulation of blastocyst.
