摘要
应用ABC免疫组织化学方法,观察了细胞外基质的主要成分IV型胶原、纤维粘连蛋白和层粘连蛋白在人卵泡的分布。结果发现:IV型胶原分布在泡膜细胞和分隔颗粒细胞、泡膜细胞的基膜上,纤维粘连蛋白分布在卵泡颗粒细胞和泡膜细胞,而层粘连蛋白则分布在基膜以及基膜两侧的颗粒细胞和泡膜细胞。这3个细胞外基质成分分布规律不同,提示它们由不同细胞产生,并在卵泡发育的微环境中发挥着既互相协同又各自独特的作用,对卵泡发育及功能具有重要意义。
bjectives :To investigate the distribution of collagen IV, fibronectin (FN)and laminin(LN)Whichare the main components of extracellular matrix(ECM ) in human ovarian follicles. Design:Immunochemistry study. Settings:Endocrinology and molecular biology laboratories in Notional Research Institute for FamilyPlanning. Methods :The ovaries were removed from th premenopausal patients who underwent salpingo- oophorec-tomy.Results:This is the first demonstration of the localization of collagen IV in human ovaries. Collagen IVwas shown in th theca ce11s and basal lamina(BL)separating the granulosa ce111s and theeca ce11s.FN pre-sented in granulosa cells and theca cells. LN was in BL,granulosa cells and theca cells.The results indicatesthat collagen IV is secreted by theca cells,FN and LN by both of these cells. Collagen IV and LN are com-ponents of BL. Conclusion ECM might not only control cellular behavior such as cell attachment,migration,differen-tiations, but also might be involved in the roles of secreting steroids and maintaining the maturation of the oocyte.
