大白鼠听皮质脑组织移植初步研究(移植组织的细胞构筑和纤维联系) 被引量:1

Preliminary Study of Transplantation on Cerebral Cortex Tissue in Auditory Cortex Areas of Adult Rat: Cytoarchitecture and Connectivity of Transplant

摘要 取Wistar大白鼠15d胚胎听皮质植入到成年受体脑相应皮质的预制脑腔中。在受体动物脑内存活4~6个月后,检查结果表明移植组织的成活率达78%。移植组织内未见细胞分层,细胞呈“团簇”状排列,有髓纤维叶状分布,有类似于正常大脑皮质的大、中形锥体细胞和颗粒细胞,锥体细胞顶树突弯曲。移植组织与受体脑之间建立了“有效移植面”。CT-HRP示踪法表明移植组织与受体脑的同侧内侧膝状体,对侧听皮质相互有纤维投射。电镜观察发现移植组织内有丰富的神经毡、正常的突触结构和突触小泡。结果提示:移植组织可能已具有代替已损听皮质并重建相应靶区的神经环路。 Embryonic auditory cortex obtained from 15-day-old Wistar rat embryo was transplanted into the prefabricated hole in appropriate cerebral cortex of adult recipitent. 4—6 monthes after transplantation, By using Nissl and Weil and Golgi-Cox preparations, the results showed there were no cellular laminations within transplants. Differentiated normal neurons were similar to the medium and large size pyramidal neurons and more granule cells while curved shape was seen at the apical dentritet of pyramidal cell. Mylinatednerve fibers passed through 'effective grafting interface'. Tracing method with CT-HRP revealed that nerve fibers of transplants projected to recipitent's ipsilateral medial geniculate body and contralateral auditor(?) cortex with each other. Electronmicroscopic examination showed rich neuropil and normal synapses, belonged to Gray's Ⅰ , Ⅱtypes and spherical synaptic vesicles were seen within Gray's Ⅰ type synapses. The results suggest that the transplants could replace and reconstruct the injured auditory cortex to some degree and form appropriated connections.
