摘要
采用B淋巴细胞杂交瘤技术制备抗人喉鳞状细胞癌单克隆抗体(McAb),经筛选得到4株分泌McAb的杂交瘤细胞株,全部收集到腹水型McAb,并检测其特异性及生物学特性。结果该McAb能与标准喉癌细胞株抗原特异性结合,抗体滴度≥1:1280倍,且能特异性地检测喉癌患者癌组织中的喉癌抗原;此McAb纯度高,分子量在100~200kD之间。单克隆杂交瘤细胞染色体均为多倍体。
The monoclonal antibody (McAb)to laryngeal squamous carcinoma was prepared by us-ing the technique of B lymph cell hybridomas, After screening,4 strains of hybridomas which can se-crete McAb were obtained and the McAbs in ascites were all collected Its specificity and biological char-acteristics were also studied.The results showed that the McAbs can combine with the antigens of stan-dard laryngeal cancer cell strain;the antibody titre were higher than l:1280;the McAbs can specifical-ly detect the laryngeal cancer antigen in carcinoma tissues; the McAbs were of high purity and the molecular weight was l0O-20kd;the cellular chromosomes of the monocfonal hybridomas were all poly-ploid.
