固体表面化学发光法测定辣根过氧化物酶的研究

Determination of HRP on the solid surface chemiluminescence technique

在不渗透阻挡滤纸上，建立了Ｌｕｍｉｎｏｌ－Ｈ２Ｏ２－ＨＲＰ固体表面化学发光分析法．游离ＨＲＰ测定的线性范围为６～８０００ｐｇ，检测限为０．６６ｐｇ／斑点，测定的相对标准偏差为７．４％（８ｐｇＨＲＰ，ｎ＝１１）．对标记的ＨＲＰ测定结果与ＥＬＩＳＡ法相一致．

A solid surface chemiluminescence method using the anti-permeate barrier as a solid substrate for the determination of HRP (Horseradish peroxidase) based on Luminol -H2O2-HRP system is described. The linear range for free HRP is 6～8000 pg. The detection limit is 0. 66 pg/spot, and the relatve standard deviation for 8 pg HRP is 7. 4% (n=11). This method has been used for assaying three labeled-HRP complexs and the results is comparable with that of ELISA.
