摘要
应用免疫组化及分子杂交方法研究了10例骨巨细胞瘤及4例胎儿骨标本。大部分多核巨细胞(MGC)及约20%~40%的单核基质细胞(STC)对抗巨噬细胞单抗标记呈阳性,胎儿骨中破骨细胞对抗巨噬细胞单抗标记亦呈阳性。在抗p53蛋白单抗染色中,30%~60%的STC标记阳性。MGC呈两种标记反应,体积大、核多的MGC呈阴性,体积小、核少的MGC呈阳性。DNA分子杂交发现有1例GCT存在部分基因片断缺失。
en giant cell tumor of bone(GCT) and four fetal bone
specimens were studied with im-munohistochemical and DNA hybrid
methods.The results showed most of the nultinuclear giant
cell(MGC),about 20%~40%mononuclear stromal cell(STC)and the
osteoclast in fetal bone showed strong positive reaction to
anti-macrophage MoAb staining.There were two types of MGC manifesting
different reactivity to anti-p53 protein MoAb staining.The dimension
of the negative staining MGC was much larger and the number of nuclei
there in was several times more than that of the positive ones.Also
there were about 30%~60%of STC that were positive to an-ti-p53
protein MoAb lebal.It was supposed that the positive reaction MGC and
STC to anti-p53 protein MoAb lebal were truly tumor elements of
GCT.In southern blot analysis,part deletion of p53 gene was detected
only in one specimen.
