摘要
采用DNA和RNA的斑点杂交分析方法,对32例乳腺癌和相应的癌旁正常组织中GST-π和int-2基因的DNA扩增和RNA转录表达情况进行研究,发现GST-π在乳腺癌中存在基因扩增和明显的mRNA表达增强,GST-π基因表达调控主要在转录水平进行;癌基因int-2与乳腺癌的发生有一定关系。同时还发现乳腺癌中GST-π与int-2基因之间存在有共扩增和共表达现象。
To further characterize the alteration of GST-pi gene expression during progression of breast cancer,both DNA and RNA levels of GST-pi in 32 cases of breast cancer and their corresponding normal tissue were examined using Biotin labelled GST-pi cDNA probe and dot hybridization. The DNA hybridization resulted in the fact than the copies of GST-pi gene altered during the progression of breast cancer. The copies of GST-pi gene were found to be elevated in 14 out of 32 cases. These proved GST-pi gene has been amplified.An increased RNA expression was also found in 30% of the cases by RNA hybridization. As to int-2 oncogene,the amplification of DNA and high expression of mRNA were determined in 5 cases with the rate of 15.6%and 10 cases with the rate of 31.3%respectively. Moreover,we revealed that GST-pi gene and int-2 gene showed obviously co-amplification and expression.
