反相高效液相色谱法测定血中阿米替林及其代谢产物

Determination of amitriptyline and its metabolites in serum by reversephase high performance liquid chromatography

摘要 用带有254nm紫外检测器的高效液相色谱仪,反相色谱法测定血清中阿米替林及其代谢产物去甲替林,E-10-OH-阿米替林,E-10-OH-去甲替林和N-去甲基去甲替林浓度。方法为碱化血清,以乙醚提取后挥干,残留物溶解后注入国产色谱柱(YWG-C_(18)) 。用盐酸赛庚啶做内标。各测定物的日内回收率为89.2-110%,变异系数小于8.8%。日间回收率为95.1-111%,变异系数小于9.6%。最低检测限为2ng,血清中最低有效测定浓度为20ng/ml。本法适用于科研或临床监测血药浓度。 A reverse-phase HPLC in conjunction with a 254 nm UV detector and a YWG-C18 column for the simultaneous measurement of the serum concentration of amitriptyline and its metabolites-nortriptyline, E-10-OH-amitriptyline, E-10 -OH-nortriptyline and N-desmethyl-nortriptyline is presented.The method involves a single extraction of alkalized serum with ether and subsequent ehromatography of the residue on the column. Cyproheptadine was used as the internal standard. Detection limits and the minimum serum concentrations of the drugs were 2 and 20 ng/ml respectively. The recoveries and C.V. of within-day and between-days were 89.2-110% and <8.8%, 95.1-111% and <9.6% respectively. It is a reliable method both for research and clinical practice.
