摘要
本文用DE—52纤维素和凝胶过滤法将猪眼晶状体中的醛糖还原酶纯化提高700倍,比活性为3220U/mg蛋白,SDS—聚丙烯酰胺凝胶电泳显示一条区带。体外实验证明,芦丁单体对纯化了的猪晶状体醛糖还原酶有显著抑制作用,ID_(50)=16.7μM。
Aldose reductase ( AR ) extrated from pig lenses was purified about 700 -fold by means of repeated gel filtration. Its specific activity was 3220 u/ mg protein. Electrophoresis of the purified AR revealed a single band. In vitro experimentation discovered that pure RUTIN ( isolated from Sophora japonica L. ) could effectively inhibit the enzymic activity of AR isolated from pig lenses and the ID50 was 16.7 umol/L.
