摘要
本实验采用高压氧下组织体外标记程序外DNA合成(unscheduledDNAsynthesis,简称UDS)放射自显影检测法,体内给药后取组织于体外标记。对4NQO、AFB1、Benz、MBN、DMBA、DEN在Wistar大鼠不同组织中诱导的UDS进行检测的结果表明:此6种化合物均呈阳性,UDS出现部位比肿瘤靶器官广泛,靶器官的UDS强度不一定比非靶器官大,UOS的出现有相对的器官和细胞特异性,明显的UDS常见于肺、肾、肝、肾上腺、胰腺和颌下腺等组织。本研究使人们对UDS发生部位与肿瘤靶器官的关系有了更多的认识,不支持某些学者认为的UDS部位仅见于肿瘤靶器官的观点。
Using
autoradiograph for detection of Unscheduled DNA synthesis(UDS)in-duced in different tissues
of Wistar rats、was examined after administration in vivo of 4NQO,AFB 1,Benz,MBN·DMBA
and DEN. The rcsult suggested that all thesechemicals are posi-tiveinducers of UDS.The sites
of UDS were more extensive than that of tumor target or- gans and the intensity of UDS in
target organs did not higher certainly than non- target or-gans.There was relative organ and cell
specificity in sites where UDS induced. The signifi-cant UDS was often encountered in
lung,kindey,liver, adrenal gland,pancreas and submaxil-lary gland, as well as in some other
tissues. This study provides more knowledge a bout therelationship between U DS sites and
target organs of tumorigenesis and did not support theviewpoint that UDS sites could belooked
upon exclusively tumor target organs。
