摘要
在火力发电厂下风侧居民区内收集大气颗粒物,经二氯甲烷提取,减压浓缩,橄榄油溶解作为受试物。给小鼠ip不同浓度受试物,观察精子畸形率,睾丸染色体畸变率和早期精细胞微核率的变化,以评价提取物对雄鼠生殖细胞的遗传毒效应。结果表明:5ml/kg抽取物对雄鼠生殖细胞无明显诱变性;10mg/kg以上可诱发精子畸形率显著高于阴性对照;25mg/kg以上提取物可使精原细胞和初级精母细胞染色体畸变率明显升高,早期精细胞微核率增加。实验结果表明大气飘尘的有机提取物可透过血睾屏障进入雄性生殖细胞,诱发遗传物质突变。
Fallout particulates from ambient air were collected in an
residential area downstream to the wind of a power plant.Then , extracted with
dichloromethane,concentrated andredisolved in olive oil.The extract was administrated ip to
mice at various dose. Sperm abnor-mality ,chromosome aberration of spermatogonia and
primary speratocyte and meiotic mi-cronuclei of early spermatids were examined.The results
showed that the extract in dose of5mg/kg exerted no significant mutagenicity on germ cells of
male mice. The extract in doseon l0mg/kg and above induced sperm abnormality with
frequency significantly higher thanthat of control;25mg/kg and above increased spermatogonia
and primary spermatocyte chro-mosomal aberration and micronuclei frequency in early
spermatids. These results indicatedthat the component of extract of fallout particulate can
permeate blood-testis barrier intogerm cells of male mice and induced mutation on genetic
material.
