脑胶质瘤c─myc癌基因重排与MDR1基因的变化 被引量:1

REARRANGEMENT OF C-MYC ONCOGENE AND AL TERATION OF MDRI GENE IN CEREBRAL GLIOMAS

摘要 作者对22例脑胶质瘤C─myc癌基因和MDR1基因Southern分析发现有5例胶质瘤C-myc癌基因发生基因重排,其中2例伴有重排序列的扩增。这些标本同时出现MDR1基因限制片段拷贝量的倍增(EcoRI,6.6kb),其倍增是脑组织2~28倍。这一分子事件不仅反映C─myc癌基因与脑胶质瘤的恶性程度和预后关系密切,而且也与胶质瘤的耐药性有关。 Genomic DNA from 22 glioma speci mens were analvsed bv sou-thern blot hybridization with both c- myc DNA and MDRI cDNA probes. The rearrange-ment of c-myc oncogene appeared in 5 cases(23%),in which 2 specimens were of therearrangemental fragment amplification. Besides, these gliomas with c-myc gene rearrarn-gement also demonstrated the restiction fragment(EcoRI,6.6 Kb) amplific ation of multi-drug resistance gene(MDR 1).Their copies were 2 to 28 folds that of normal cerebraltissue specimens. These molecular events showed c-myc rearrangement and amplificationnot only related to the malignant grade and prognosis of human gliomas,but also tothe alteration of MDRl gene in gliomas.
