摘要
维吾尔族宫颈癌组织中H-ras癌基因312bp含第12位密码于的DNA顺序经聚合酶链反应体外扩增,结合限制性内切酶MspI限制片段长短的多态现象分析,以检测第12位密码子突变,结果在53例官颈癌中只检出1例存在第12位密码子突变,在20例慢性宫颈炎或宫颈糜烂中无1例检出。说明H-ras癌基因第12位密码子突变可能不是维吾尔族官颈癌中癌基因激活的主要方式。
The 312bp DNA sequence con atining codon 12
of H-ras oncogenein Uygurian cervical carcinoma was amplified by the
polymerase chain reaction(PCR),andthen the restriction fragment
length polymorphisms of the amplified sequence was analy-zed using
the restriction endonuclease Msp I to detect mutations at codon
12.Point mu-tations at codon 12 were found in 1 of 53 Uygurian
cervical carcinomas. There were nopoint mutations at codon 12 in any
of 20 patients with chronic cervicitis and cervicalerosions. These
observations sugges that H-ras gene mutations at codon 12 may not
bethe main oncogene activation mechanism in Uygurian cervical
carcinoma.
