U_（937）细胞c──fos癌基因限制性多态性分析

Restriction fragment length polymorphism of c──fos gene in U937 cells

本文分别用ＥｃｏＲＩ、ＨｉｎｄⅢ、ＢａｍＨⅠ、ＨａｅⅢ限制性内切酶对Ｕ９３７细胞基因组ＤＮＡ和正常人基因组ＤＮＡ进行Ｓｏｕｔｈｅｒｎｂｌｏｔ分析，结果表明Ｕ９３７细胞存在Ｃ──ｆｏｓ基因限制性多态性改变，并伴随着基因扩增。

The genomic DNA from U937 cells and normal human PMNCs was analysed throughSouthern blot with the restriction endonuclease EcoRⅠ,Hind Ⅲ,BamH Ⅰ,Hae Ⅲ.The results showedthat there was a novel band in U937 cells compared with the normal human genome,and strength of theband was bigger than that of normal genome. These data indicate that there are structural changes andamplification of c──fos gene in U937 cell line.
