抗独特型抑制法测定可溶性IL一2受体的实验研究

The measurement of sIL-2R by anti-idiotype inhibition ELISA

本研究用抗独特型抑制法测定可溶性ＩＬ一２受体（ｓＩＬ一２Ｒ）。主要步骤为：以抗ＩＬ一２Ｒａ单抗５Ｇ１包被，加入ｓＩＬ一２Ｒ及针对包被单抗的抗独特型酶标抗体ＨＲＰ一２Ｇ３。ｓＩＬ一２Ｒ与ＨＲＰ一２Ｇ３竞争结合包被单抗，其含量与显色ＯＤ值成反比。本法简便，灵敏，特异性好，可望用于基础及临床研究。

In this work,anti-idiotype inhibition ELISA was used to measure soluble IL-2 re-ceptor(sIL-2R).The main procedures were as follows :Microplate was coated with anti-IL-2Ramonoclonal antibody(McAb)5G1;after blocking with l% BSA,sIL-2R and HRP-2G3 which is ananti-5G1 idiotype McAb were added simultaneously. In this system,sIL-2R could inhibit the bind-ing between 5Gl and HRP-2G3,and there was a negative correlation between sIL-2R and OD492value. The results show that this method is spccific and sensitive for measurement of sIL-2R.
