PCR技术检测儿童急性淋巴细胞白血病TcRγ基因重排

摘要 应用PCR技术检测20例急性淋巴细胞白血病TcRγ基因(VγI,Jγ_(1-2))重排,其扩增产物的长度为400bp左右。初治和复发的13例患儿中有7例检出TcRγ基因重排,其中复发的3例患儿均有TcRγ基因重排,完全缓解的7例患儿中有4例检出TcRγ基因重排,此4例患儿目前继续维持治疗,尚无复发之临床表现。TcRγ基因重排几乎存在于全部的T-ALL和大部分B-ALL中,其稳定性强。对TcRγ基因重排的检测可用于协助急淋患儿的诊断及长期监测。进一步研究对指导治疗,判断预后和发病机理的研究有重要意义。 Abstract We detected the T-cell receptorγ gene rearrangments in 20 children with acutelymphoblastic leukemia by means of PCR method. The band of products is about 400bp inlength.7 patients were gene rearrangment positive in 13 nonremission patients. In all 3 clinicallyrelapsed patients, we found TcR γ gene rearrangment. We detected TcR γ gene rearrangmentpositive in 4 of 7 complete remission patients. These 4 patients have not the sign of relapse tillnow. The rearrangment of TCR γcould be found in almost all T-ALL and most B-ALL patients.Its stability is good.To dectect therearangment of TCR γcan help us in the diagnosis of ALLand monitor the development of the disease for a long time. The further study is very importantfor guiding the treatment and determining the prognosis and studying the mechanism of disease.
