负重力竭性游泳对大鼠血清胰岛素血糖浓度的影响及胰岛B细胞图像分析 被引量:10

Effects of Exhaustive Swimming with a Load Attached to the Body on Serum Insulin and Blood Glucose Concentration and the Analysis of Islet B Cell Image in Rats

摘要 采用放免分析法及氧化酶法对大鼠负重游泳至力竭状态时血清胰岛素(Ins)及血糖(BG)水平进行测定,同时利用免疫组化PAP法分别显示其胰岛Ins-和IAPP-(胰岛淀粉样多肽)免疫反应细胞,通过图像分析方法对实验组及对照组大鼠胰岛免疫反应细胞的面积进行测量。结果显示,力竭状态下大鼠血清Ins↓BG↑(P<0.01)。这种变化是由于胰岛B细胞释放Ins受到抑制所致。同时发现,以往认为从B细胞相伴释放的Ins与IAPP的分泌并非完全一致。认为其差异乃是由于B细胞内存在着IAPP/Ins不同比例的分泌颗粒随机体的能量代谢需要而释放的结果。 Serum insulin (Ins) and blood glucose (BG) level of rats were determined by radioimmunoassay (RIA) and oxidase method when they swam to exhaustion with loads attached to their bodies.The islet Ins and IAPP (islet amyloid polypiptide) immunoreactive cells were shown by immunohistochemistry PAP method. And the areas of the islet immunoreactive cells in the experimental rats and the control rats were measured by image analysis method. The results showed that their Ins reduced and BG increased (P<0.01) during exhaustion. Those changes resulted from the inhibition of the Ins released from the islet B cells. In the meantime,it was found that the secretion of Ins was not the same as the secretion of IAPP,which were thought to be accompanying released from the B cells. It is suggested that the difference between them is because in the B cells there are different ratio of IAPP and Ins secretory granules,which are released with the demand of the energy metabolism of the body.
