etection of citrus exocortis viroid(CEV)was conducted by using photobiotin -labelledF20 probe. Dot hybridization indicated hat F20 was very specific to CEV sequence, and thatthe highest dilution of crude CEV-nucleic acid extracts and the minimal amount of CEVdetected was about 1/100 and 1ng rtspectively. The concentration of photobiotiny-labelledprobe for dot hybridization should be at least 0.5μg/ml in general. The blocking reagent Tween 20 can effectively clean the membrane background and enhance its sensitivity.
