摘要
以兔出血症病毒(RHDV)德国株基因组cDNA的部分片段为探针,与中国的NJ株和LW株RHDV的基因组做分子杂交,结果表明,它们属于同一病毒。将中国株RHDV感染兔肝组织,用硫氰酸胍-氯化铯超速离心法提取的核酸沉淀,或将提纯病毒悬液经苯酚抽提法获取的核酸沉淀,在用RNase或DNase,消化后直接做Dotblot,或将相应样品做PCR后再做Dotblot或Southernblot,再与经Digoxigenin标记的cDNA探针做分子杂交,结果证明,在中国的二个不同来源的兔出血症病料中,与德国株RHDV基因组cDNA同源的病毒是一种ssRNA病毒。本文还讨论了在中国兔出血症病料中存在另一种DNA病毒的可能性。
ith labeled cDNA clone of German RHDV strain as the probe,dot blot and Sorthem blot of RNase-digested and DNase I-digested nucleic acid samples from infceted rabbit liversuspensions extracted by granidine isothiocyanate-caesium chloride gradient centifugation orfrom purified virus suspension by phenol-extraction were conducted.Dot blot and Southem blot of PCR products from the above nucleic acid samples were also carried out .The resrlts demonstrated that the genomes of two Chinese strains (NJ and LW)of RHDV werehomologous with the cDNA from the German RHDV genome and were ssRNA
