摘要
用组织培养方法筛选获得羊草(Aveurolepidiumchinense(Trin.)Kitag.)抗羟脯氨酸(HYP)变异系HR20一8。该变异系统细胞内游离氨基酸和蛋白质中氨基酸含量均发生了较大的变化,与供体对照比较,分别提高2.35倍和1.40倍。其中的脯氨酸组分在变异系体内表现异常增加,游离和蛋白质中的脯氨酸组分分别提高6.6倍和3.0倍,对脯氨酸合成途径中的γ一谷氨酸激酶的特性分析表明,变异系HR20一8细胞内γ一谷氨酸激酶的活性比供体提高2.5倍。
Avariant(HR20一8)resistant to hydroxyproline was obtained from Aneurolepidium chinense through tis-sue culture. It was observed that there were significant increases in the content of free amino acid and thatof protein amino acid in cells of the variant:by 2.35 and 1.40 times higher,respectively,in comparisonto the levels in the donor plant.The increase in proline content was particularly notable:by 6.6 and 3.0times higher for free and protein amino acids respectively.An examination of γ一glutamyl kinase involvedin the process of proline biosynthesis indicated that the activity of the kinase in HR20一8 was 2.5 timeshigher than that in the donor plant.
