摘要
本实验以24只Wistar大鼠的密度梯度离心后的周血淋巴细胞作为观察对象,应用免疫组织化学方法显示EGFR—IR和ACTH—IR,用生物素标记的cRNA探针原位杂交显示EGF的基因表达,此外对比观察了PHA刺激转化的淋巴母细胞和未转化的淋巴细胞的ACTH-IR。结果见到淋巴细胞EGFR-IR呈现阳性、EGF-mRVA信号呈蓝紫色颗粒布满胞质,提示淋巴细胞对EGF可具有自分泌甚至旁分泌效应。ACTH-IR在未转化淋巴细胞呈弱阳性而转化的淋巴母细胞显著增强。
In this paper the EGFR-IR and EGF gene expressioll have been studioic by theimmullohistochemistry technique and in situ hybridization witch the salllples of rat circuirtinglymphocytes. Besides. the ACTH -IR was contrasted between transfomed and nontrans-formed lymphocytes. The results indicated that the EGFR-IR appeared positive reactloll andEGF-mRNA signal locallied at the lymphocytes. It suggests that there was EGF effect inlymphocytes via autocrine or paracrine rout. The ACTH-IR in the nontransformed lympto- cytes appeared week positivity. whereas, the ACTHI-IR was markedly enhaneed in the trans- formed lymphocytes.
