固蓝B法测定血清胆红素的改进

IMPROVED FAST BLUE B SALT METHOD FOR MEASUREMENT OF BILIRUBIN IN SERUM

报告对固蓝Ｂ单一试剂测定血清总与结合的胆红素法的改进，在联合应用甲醇和尿素作总胆红素显色的加速剂，降低了固蓝Ｂ的浓度。总胆红素的回收率为９５．２～１０２．２％，批内重复性，总胆红素ＣＶ＝１．２％，结合胆红素ＣＶ＝２．７％；批间重复性，总胆红素ＣＶ在２．３～３．８％之间，结合胆红素为４．０～４．３％。本法（ｙ）与Ｊ—Ｇ法（ｘ）比较，总胆红素两法密切相关（ｒ＝０．９９８９ｒ＝０．９８５１ｘ＋１．０６７６），结合胆红素，以Ｊ—Ｇ直接法定值的黄疸血清作参考物测得的结果表明两法紧密相关（ｒ＝０．９９９７，ｒ＝０．９９７ｘ＋０．５６６７），但以未结合胆红素作标准则有明显的比例偏差（ｒ＝０．９９３０，ｙ＝０．８４６６ｘ—３．５０７１）。

The modified fast blue B salt method for determination of total andconjugated bilirubin in serum were described。 Within-run precisions(CVs)of total and conjugated bilirubin were 1.2%and 2. 7%, respectively.Between- run CVs were 3. 8% 2.3%at the total bilirubin concentration of 21.26 121.07 μmol/L and 4.3% 4. 0%at conjugated biliubrin 38.82 59.85μmol/L.The recovery of total bilirubin was 95。2% 102.2%。The linear range of the method reached up to at least 256 μmol/L(15mg/dL)for both total and conjugated bilirubin.Results obtained by the describedmethod(Y) closely correlated with those by modified Jendrassik-Grof( J-G) method(X):r=0.9989,y=0.985x+1.0676pmo/L for total bilirubin;R=0.9997,y=0.997x+0.5667μ mol/L for conjugated bilirubin determined by using pool jaundice serumcalibrated by modified J G direct method as reference material。
