摘要
利用3段分别代表HEV3个基因区的合成多肽,研制成ELISA抗HEV检测试剂。此试剂对IgG及IgM检出阳性率在急性NANBNC肝炎患者中分别为54.1%(53/98)和33.7%(33/98),在其他疾病患者及正常献血员中则分别为2.7%~6.3%和0%~2.6%。与Genelabs重组抗原生产的检测试剂比较,两者有很好的一致性,其阳性符合率为90.0%,阴性符合率为98.5%,总符合率为97.4%。对于急性戊型肝炎患者动态监测,7~20d内大部分患者能检出抗HEVIgG或IgM。这些结果表明,利用HEV合成肽能特异地检测戊肝病毒感染者的抗HEVIgG及IgM,有助于临床诊断及流行病学调查。
Three synthetic peptides representing distinct antigenic epitopes from three genomic regions of hepatitis E virus(HEV) were used to develop ELISAs for detecting antiboclies to HEV. Both IgG and IgM antitodies to HEV were detected respectively in 53 of 98(54.1%) and 33 of 98(33.7%)sera of acute NANBNC hepatitis.Very fewIgG (2.7 % ~ 6.3 % )and IgM (0% ~ 2.6 %) antibodies were detected in sera from the patients with other diseases and normal blood donors. This assay was in good agreement with the Genelabs kit utilizing recombinant HEV antigens with the positive coincidence of 90.0%,negative coincidence of 98.5% and total coincidence of 97.4% in 982 serum slpcimens.The IgG and IgM class antibodies were detected in most cases (5/6) within 7 to 20 d after the onset of hepatitis E.Our data indicate that the method for detection of HEV antibodies based on synthetic peptides is also very specific,sensitive and usable for clinical practice and epidemiologic investigation.
