组胺H＿1受体拮抗剂特非那定对正常和致敏豚鼠气管螺旋条的作用

A study on the effect of terfenadine, a histamine H_1 receptor antagonist,on normal and sensitized guinea-pig tracheal spirals in vitro

为了探讨组胺Ｈ１受体（Ｈ１Ｒ）在哮喘发病中的作用，我们研究了特非那定对豚鼠离体气管螺旋条的作用。２４只豚鼠随机分成２组，对其中一组予卵蛋白致敏。取其气管制成螺旋条，以组织浸浴法研究特非那定的作用。结果显示，１×１０－６和１×１０－７ｍｏｌ／Ｌ特非那定可抑制组胺诱发的气管螺旋条收缩、但不能使已收缩的螺旋条舒张，特非那定对组胺诱发致敏螺旋条收缩的抑制作用弱于非致敏螺旋条，提示致敏组织的Ｈ１Ｒ功能亢进或Ｈ２Ｒ功能低下，特非那定可用于哮喘预防性治疗。

To investigate the effect of histamine H_1 receptor(H_1R) on pathogenesis in asthma,we studied the effect of terfenadine on guinea-pig spirals in vitro.Twenty-four guinea-pigs were divided into two groups,one of them was sensitized with ovalbumine.The responses of the spirals to histamine and terfenadine were studied with tissue bath experiment.The results showed that(1)the dose-response curves of the spirals to hsitamine were shift to right in adding terfenadine prior to histamine and terfenadine had not any relaxation effect on contracted spirals;(2) the inhibition of terfenadine was weaker in the sensitized group than that in the normal group, suggesting that the reactivity of H_1R might be higher or H_2R lower in sensitized animals.Terfenadine might be used for prophylaxis of asthma.
