苦参碱对巨噬细胞释放肿瘤坏死因子及其蛋白激酶C活性的影响 被引量:58

摘要 研究了用苦参碱(Mat)处理内毒素诱导卡西霉素启动的小鼠腹腔巨噬细胞(M ),观察其对分泌肿瘤坏死因子(TNF)的影响,及在此过程中M  胞浆和胞膜部分蛋白激酶C(PKC)活性的变化。结果表明:Mat0.5、1.0mmol/L均可显著抑制TNF分泌,对M胞浆和胞膜PKC活性也有抑制作用。提示:Mat抑制TNF分泌与其抑制M 内PKC活性有关。 Studies were designed to determine the effect of matrine on the induction of tumor necrosis factor(TNF) production as well as the change of protein kinase C(PKC) activity in cytosol fraction and membrane fraction during the induction.Matrine 0.5,1.0 mmol/L markedly inhibited lipopolysaccharides induced TNF release from peritoneal macrophages (M) primed by calcimycin(1μmol/L) and PKC activity in cytosol fraction and membrane fraction of M.These results suggest that inhibitory effect of matrine on TNF production is possibly contributed to its inhibitory action on the intercellular PKC activity.
