γ-射线诱发人淋巴细胞微核形成与细胞周期关系的初步研究 被引量:8

摘要 本研究用γ-射线诱发人外周血淋巴细胞微核(MN),使用细胞周期控制、BrdU免疫荧光染色、松胞素B(CB)阻滞胞质分裂等技术,研究人淋巴细胞微核形成与细胞周期间的定量关系。结果:①G0期和G1期淋巴细胞照射组和对照组相比,微核率(MNF)和微核细胞率(MNCF)均无明显变化(P>0.05)。②S期和G2期淋巴细胞照射组和对照组相比,MNCF均明显增加(P<0.01)。(3)M期淋巴细胞照射组与对照组相比,MNF与MNCF分别增加90多倍和70多倍。结论:在人淋巴细胞周期中S期和G2期可形成MN,但M期是形成MN的主要阶段。 The quantitative relationship of cell cycle with the micronucleus formation of the lymphocytes of human peripheral blood induced with gamma ray irradiation were studied with the control of cell cycle,bromodeoxyuridine(BrdU)immunofluorescent staining and blockade of cytoplasm division with cytochalasin B(CB).It was found that 1.The micronucleus frequency(MNF)and micronucleated cell frequency(MNCF)of C10 and C11 phases of the irradiated lymphocytes were not significantly different from those of the control(P>0.05);2.The MNCF of S and C12 phases of irradiated lymphocytes was significantly higher than that of the control(P<0.01)and 3.The MNF and MNCF of M phase of irradiated lymphocytes were 90 times and 70 times higher than the control respectively,These findings suggest that a part of micronuclei can be formed in S and C12 phases of human lymphocytes and M phase is the main stage of micronucleus formation in the whole cell cvcle.
