兔胎脑皮层组织4℃保存的细胞活性变化

摘要 本实验取胎龄为23~24d的胎兔大脑皮层组织,分别以iloprost液及生理盐水为保存液,在4℃保存前后制成细胞悬液,不同浓度碘化丙啶染色后经流式细胞分析和台盼蓝排斥试验测定细胞活性。结果表明,iloprost液可以对兔胎脑皮质提供良好的保护作用,但随保存时间延长,细胞活性仍不可避免地下降,提示应进一步深入研究。最后,本研究还表明不同浓度碘化丙啶染色经流式细胞分析用于细胞活性测定较台盼蓝排斥试验更敏感、准确。 Cerebral cortical tissue from rabbit fetus of 23~24 gestation was preserved in saline or iloprost solution(50ng/ml)at 4 ℃.Cell suspension of the prepreserved and postpreserved cerebral cortex of rabbit fetus was prepared with mechanical method and the viability of the cells was examined with the fluorescent stain with PI of different concentrations through flow cytometry and with trypan blue exclusion,It was found that iloprost solution exerted more potent protection on the embryonic cerebral cortical tissue than saline.Nevertheless,the viability of the cortical cells was inevitably decreased when the preservation was prolonged and that the fluorescent stain with PI in different concentrations through flow cytometry was more sensitive and accurate to examine the cellular viability than trypan blue exclusion.Further investigation is imperative to elucidate the problem of cellular viability of preserved rabbit cerebral cortex.
