摘要
目的:确定血管活性肠肽(VIP)对培养大鼠肝癌细胞生长的影响.方法:采用四甲基偶氮唑盐比色法(MTT试验)和细胞计数法.结果:VIP对癌细胞增殖有明显刺激作用,加入1μmol/LVIP后,培养的大鼠肝癌细胞的数量在第3日即显著增加,第4,5日细胞数增加十分显著(P<0.01).对培养肝癌细胞生长的刺激效应随着VP浓度的增加而增强.最低有效浓度约为0.5μmol/L.VIP和肝癌细胞连续作用12h后,对培养癌细胞的生长刺激效应可维持数日.结论:VIP可显著刺激体外培养的大鼠肝癌细胞的生长.
Objective:Vasoactive intestinal peptide ( VIP ) can stimulate the growth of
a variety of tumorcells, but there is no report about the effect of VIP on the growth of hepatoma
cells. The aim of this experi-ment was to confirm the effect of VIP on the growth of cultured rat
hepatoma cells. Methods:The tetrazoli-um colorimetric assay (MTT assay) and cell counting
method were used.Results:VIP stimulated the prolif-eration of the cultured rat hepatoma FSK
7902 cells obviously. The addition of 1 μmol/L VIP caused a signif-icant increase in the
number of cultured hepatoma cells on 3rd and maximal increase occurred on 4th day and5th
day(P<0.01).The growth promoting effect was greater as the concentration of VIP increased.
Thelowest effective concentration of VIP was 0.5 μmol/L. Exposure the VIP for 12 hours
followed by removal ofthe peptide resulted in sustained growth promotion for several days.
Conclusion:VLP stimulates the growthof cultured rat hepatoma cells.
