摘要
胰脏是提取胰岛素及多种酶的原料,为提高产率及高度综合利用,作者采用酸、碱抽提、硫酸铵分级沉淀、膜过滤、色谱分离等技术,从1kg猪胰脏中同时分离纯化酶达9种之多,而各酶的比活力及总活力分别为:胰蛋白酶16000BAEEU/mg蛋白,28.4×10 ̄6BAEEU;弹性蛋白酶120U/mg蛋白,6000U;缓激肽酶40.38U/mg蛋白,3876U;胰凝乳蛋白酶22000ATEEU/mg蛋白,19.5×10 ̄6ATEEU;胰蛋白酶抑制剂2089T.I.U/mg蛋白,31727T.I.U;羧肽酶A9.5U/mg蛋白,3988U;羧肽酶B154.8U/mg蛋白,34675.2U;核糖核酸酶A80KunitzU/mg蛋白,800U;脱氧核糖核酸酶1137.8U/mg蛋白,106000U,本法经多次优选优化而成,建立了简易合理的工艺流程,为充分利用资源,降低成本,建立工业化生产开辟了新的途径.
Pancreas is a source of
insulin and many enzymes. Further exploitation and increasing consumption have resulted
in its shortage. Therefore, how to raise its yield and how to make a multipurpose use of it have
become a significant job. In the present experiment,employing such techniques as Acid &
alkaline extrac-tion,Ammonium sulfate fractional precipitation,Membrane infiltration and
Chromatographic separation the authors simultaneously extracted and purified as many as 9
kinds of enzymes from 1kg of porcine pancreas. Their specific and total activities were as
follows each: trypsin 16000 BAEEU/mg prot.,28.8×10 ̄6 BAEEU;elastase 120 U/mg prot.,6000
U; kallikrein 40 U/mg prot.,3876 U; chymotrypsin 22000 ATEE U/mg prot.,19.5×10 ̄6 ATEE U;
trypsin- inhibitor 2089 T. I. U/mg prot.,31727 T.I. U; CpA 9. 52 U/mg prot.,3988U; CpB 154. 8
U/mg prot.,34675U;RNase A 80 kunitz U/mg prot.,800U;DNase 1137. 8 U/mg prot.,106000 U.
The procedure underwent several optimizations, thus established a rational and simplified
technological process,which promises a new way of full utilization of pancreas and industrial
production of variety of enzymes from it at low cost.
