摘要
用粘附式细胞仪(ACAS570)检测了培养细胞缝隙连接通讯功能。预先用荧光探针6-羟基荧光素乙酰乙酸盐进行细胞染色,然后光漂白细胞中的荧光分子。由于邻近细胞中未漂白的荧光分子通过缝隙连接扩散到漂白的细胞内,故可监测漂白细胞荧光强度的恢复。根据荧光恢复的比率,了解缝隙连接通讯功能的状态。

newmethod for the measurement of gap junction in cultured cells was reported.The method was
based on thefluorescence intensity reduction of the photobleached cells(the cells were
previously stained by a fluores-cent probe,6-carboxy-fluorescein diacetate CFDA)and its
recovery through gap junction-mediate fluores-cent molecule diffusion from adjacent
unphotobleached cells.According to the fluorescence recovery rate,the function of gap junction
communication could be evaluated.
