摘要
以HL_(60)、U_(937)细胞系为体外模型,应用透射电子显微镜技术,观察了重组人转化生长因子β_1(rhTGF-β_1)对以上两种人白血病细胞系超微结构的影响,结果显示:U_(937)细胞对rhTGF-β_1的反应比HL_(60)细胞敏感,经rhTGF-β_16ng/ml作用4天后,U_(937)细胞之胞核不规则,核仁减少或消失,出现较多的粗面内质网,胞浆中可见吞噬小体,进一步证实rhTGF-β_1可诱导U_(937)细胞向成熟分化,分化途经仍为单核巨噬细胞系。
eukaemia cell line(HL_(60),U_(937))was
used as an in vitro model in our study. We investigated the effectsof
rhTGF-β_1 on ultrastructure of the two cell lines with transmission
electron microscopy. The results showthat the susceptibility of
U_(937) to TGF-β_1 was higher than that of the HL_(60) cell.Four
days after the inductionof U_(937) cells by TGF-β_1 6ng/ml the size
of individual cells was enlarged and the shape of nuclei was
irregu-lar.Decrease in number or disappearance of nucleoli was noted.
The rough endoplasmic reticula increasedmarkedly in number,There were
phagocytic bodies in the cytoplasm. The findings of the present
sttidydemonstrate that U_(937) cells could differentiate into
mononuclear macrophages following the induction byTGF-β_1.
