摘要
本文采用限制性核酸内切酶技术,对本室分离的Q热立克次体七医株,进行了DNA酶切片段的电泳分析,并以国外标准株(Grita株)作为参照。Q热立克次体经泛影葡胺密度梯度离心法进行分离纯化。提纯的立克次体DNA,用限制性核酸内切酶(HaeⅢ和BglⅠ)分别消解后,再经琼脂糖凝胶电泳法分析。本文结果证实,七医株和Grita株的酶切电泳图谱存在差异。这种差异在HaeⅢ酶谱上尤为明显。不仅两株立克次体DNA的绝大多数酶切片段的迁移距离不同,而且Grita株明显比七医株缺少包括4.3kb在内的几个HaeⅢ片段。
The preparation of DNA from Qiyi strain is analysed by using restriction endonuclease technique. Another foreign standard strain (Grita strain) is used as the contrast. Coxiella burnetii are purified via the diatrizoate meglumine density gradient precipitating procedure.The neat DNA are digested with HaeIII or BgII individually, following with an analysis with agarose gel electrophoresis. This study shows that the restriction patterns of Qiyi and Grita strains are different.The difference is quite distinct on Haelll electrophoretogram. Not only do distances migrated by most restriction fragments from Qiyi and Grita strains be distinct, but also some. Haelll fragments involving 4.3 Kb are absent in Grita strain.
