摘要
本文通过FACS(荧光激活细胞分选器)技术,利用单克隆抗体RA3-6B2抗Ly-5(B220)研究了Ly-5(B220)在新制备的小鼠脾脏B细胞上的表达,特别是用不同的丝裂原刺激而激活B细胞后Ly-5表达的变化,发现脾脏 B细胞形成两个主要细胞群,浮力密度较高的小B细胞高度表达 Ly-5(Ly-5hi),浮力密度较低的大B细胞则为Ly-5low。用多克隆刺激因子LPS(细菌脂多糖)刺激脾脏高浮力密度小B细胞,可诱导ty-5的表达降低。单独用膜交联剂抗-IgM刺激,ty-5的表达仍保持较高状态,但当用抗-IgM与白细胞介素-6一起刺激时,则诱导Ly-5的表达降低。这些结果表明,B细胞活化后Ly-5的表达降低,Ly-5low可作为B细胞活化的一种标志。
The expression of Ly-5 (B220) in splenic B cell of mouse and the change of Ly-5 (B220) during B cell activation were studied by using FACS technique. The results showed that the splenic B cell form two populations. The smaller splenic B cell with high-density has the phenotype of Ly-5hi, and the larger population with low-density is of Ly-5low phenotype. Activation of resting B cells by LPS and anti-IgM induces differences in the level of Ly-5 expression. Stimulation of resting B cell of Ly-5hi. phenotype, but the latter remains high as the anti-IgM stimulation is used alone for. Additional stimulation of IL-6 following anti-IgM stimulation can decrease the level of Ly-5 expression. However, IL-1, IL-2, or IL-4, whether alone or in various combinations, can not induce the loss of Ly-5. Taking those together, we can conclude that the expression of Ly-5 goes down following B cell activation and Ly-5low may serve as an activation marker of B cell activation.
