摘要
本文介绍了一种灵敏测定大白鼠皮下徽透析液中葡萄糖的液相色谱-电化学(LCEC)分析方法.本法用柱后葡萄糖氧化酶反应器将色谱分析柱中分离出的葡萄糖转化成过氧化氢,再用过氧化物酶电极检测生成的过氧化氢.本法中的过氧化物酶电极是将含有过氧化物酶的氧化还原聚合物涂覆在破碳电极表面制成的,可于+100 mV(υs.Ag/AgCl)测定过氧化氢的还原电流.与常规的柱后检测过氧化氢的铂电极相比,过氧化物酶电极显著提高了灵敏度、检测限和检测稳定性.
A liquid chromatography-electrochemical detection method was developed for the determination of glucose in rat subcutaneous microdialysate. The liquid chromatography is coupled with a post-column glucose oxidase enzymatic reactor for the production of hydrogen peroxide, which is in turn detected by a peroxidase electrode. The peroxidase electrode is a glassy carbon electrode coated with a redox polymer film containing peroxidase. It can be operated at +100 mV(vs. Ag/AgCl) for the reduction of H2O2. Compared with a conventional platinum electrode for the post-column detection of H2O2, the peroxidase cathode exhibits significant improvement in sensitivity, detection limit, and stability.
