摘要 在MS附加BA0.5—1.0mg·L-1、IAA1.0mg·L-1及Ad80mg·L-1培养基上建立罗汉果试管株系.分别用热处理和微茎尖进行脱除花叶病毒培育无病苗的研究.结果表明;用0.8—1.0mm微茎尖培养和用38.5℃热处理1周后再切取2mm茎尖培养,脱毒效果最好,其脱毒率和成活率均达100%.采用植物形态特征比较法、黄瓜作指示植物的生物学鉴定法及电镜观察法进行了小苗脱毒鉴定.研究还对带毒苗与经鉴定为脱毒苗的叶片氨基酸进行测定,结果显示两者间游离氨基酸含量差异显著. Meristem tip culturing and heat therapy were used in various cultivars of Siraitia grosvenorii (Swingle) C. Jeffery fot rapid clonal propagation of their mosaic disease free plants. Both meristems and heat treated meristems were cultured on MS medium containing 0.5 1.0 mg·L-1BA, 1.0 mg·L-1 IAA and 80 mg·L-1. Plants derived from both 1 mm long meristems and 2 mm long heat treated (38. 5℃, 1 week) meristems were 100% free from disease, as determined by visual inspection, mechanical inoculation to Cucumis sativas, and electron microscopy. The determination of aminoacid contents of leaves showed that there were significant differences between the disease and disease free plants.
