摘要
采用最常用的3种切片技术对枇杷的花柱、胚囊和胚乳的发育结构进行光镜、电镜观察比较,结果表明:石蜡切片法最适宜于观察枇杷胚胎发主发育的基本过程;石蜡切片和半薄切片均看到住头乳突细胞,但半薄切片观察乳突细胞更清晰且容易发现传递细胞的存在;超薄切片则能更清楚地显示传递细胞的内构,包括细胞器沿壁内脊附近分布、核瓣裂等特点.半簿切片和超薄切片均证实了花柱管胞的存在及其形态学特征.用石蜡切片易观察到整体胚囊结构并可看到助细胞丝状器,半薄切片很难对胶囊进行全面观察,但能更清楚地显示搜捕到的丝状器及其周边结构;超薄切片因胚囊结构定位不易而难以观察.石蜡切片、半薄切片和超薄切片均可用在胚乳发生发育的观察上,且三者有一定的互补性,综合应用,效果较佳.文中简要评述了3种切片法的优缺点及实用范围,并讨论了观察的某些重要发育结构的机制.
Using the comparative methods of paraffin-section,semithin-section with light microscopy and ultrathin-section with transmisson electron microscopy,some aspects,i.e.stigma and style,embryo sac and endosperm in embryology of loquat had been studied.The resnlts showed that there were papillos cells on the wet stigma;the style was solid with transfer cells and annular tracheids in transmitting tissue; the embryo sac conformed to the unispore-polygonum type; there existed district filiform apparatus on synergids and the development of the endosperm belonged to the nuclear type. It was considered that the paraffin-section was main method to investigate the basal proces of embryology in loquat, while it was more suitable for both semithin-section and ultrathin-section to investigate some aspects such as transfer cell and tracheids in loquat,moreover. ultrathin-section could be suitably used to study ultrastucture such as wall ingrowths and organelles of transfer cell and developmental mechanism in loquat embryology.
