利用二乙基二硫代氨基甲酸钠-血卟啉衍生物-激光抗癌的研究 被引量:2

摘要 利用二乙基二硫代氨基甲酸钠(DDC)-血卟啉衍生物(HpD)-激光抗癌的结果表明,DDC可以显著增强HpD-激光对癌细胞的杀伤作用。以HpD-激光处理,癌细胞(H_(22))的死亡率仅为15.9%,而以DDC-HpD-激光处理,其死亡率则高达92.9%。对于荷瘤(H_(22))小鼠,在HpD-激光治疗时,先给予DDC,可提高其疗效。DDC,HpD和激光在抗癌中,三者具有协同作用。 We suggest that diethyldithiocarbamate (DDC), hematoporphyrin derivative (HpD) and laser can destroy tumor cells by enhancing the free radical reactions in them. Tumor cells (H22) were suspended in RPMI-1640 medium supplemented with calf serum, and incubated at 37℃ and treated with DDC-HpD-laser, HpD-laser, DDC-laser or DDC-HpD-laser,the viability of the tumor cells was checked by trypan blue dye exclusions. Three hours after irradiation with laser, the mortality of the tumor cells treated with DDC-HpD-laser was 92.2%, whereas those of the tumor cells treated with HpD-laser, DDC-laser, or DDC-HpD were less than 30% respectively. The difference between the groups of DDC-HpD-laser and HpD-laser is very significant (P< 0.01). Tumor cells (H22) were transplanted subcutaneously in 42 mice. Three days later, half of them were treated with DDC-HpD-laser and the other half with HpD-laser. On 50th day after transplantation, 16 of the mice treated with DDC-HpD-laser survived, and 8 of them recovered from cancer, whereas the number of the survived mice treated with HpD-laser was 6, and only 2 of them recovered from cancer. The preliminary results indicate that DDC can sensitize the tumor cells to HpD-laser. The method of treating cancer with DDC.HpD-laser is therefore worth further studying.
