The Real-time Second-order Derivitive Spectral Analysis of C-terminal of hEGF by RP-HPLC

Ｔｒｐ（色氨酸）与Ｔｙｒ（酪氨酸）二阶微分光谱能被用来在线检测ＲＰ－ＨＰＬＣ（反相高效液相色谱）中含芳香族氨基酸的肽段，这些特征光谱是通过二极管矩阵检测器（ＤＡＤ）得到的，用这种方法分析人表皮生长因子（ｈＥＧＦ）的胰蛋白酶酶解图谱，能鉴别出含Ｔｒｐ的Ｃ－端肽。

The characteristic second-order derivitive spectra of tryptophan and tyrosine are described and used toidentify the peptides containing tyrosine and tryptophan residues during their elution from reversed-phaseHPLC. Spectral data were acquired with a scanning diode array detector.The method allows the non-destructive identification of tryptophan residues in tryptic maps of hEGF and enables the selection andisolation of C-terminal of hEGF.
