摘要 利用CFU-GM培养技术,探讨了Ge-132在体外对正常人CFU-GM产率的影响.结果表明,Ge-132在体外能显著增加CFU-GM产率,而且有效剂量范围大;去除贴壁细胞对Ge-132的造血刺激作用无明显影响,但除去T淋巴细胞,这种促进作用消失.把Ce-132直接加入无集落刺激活性(CSA)的培养体系中,未见集落生长.这些结果表明,Ge-132并非直接作用于粒-单祖细胞. The effect of Ge-132 on proliferation and differentiation of normal human marrow granulocyte and monocyte-macrophage progenitors ( CFU-GM ) were investigated. We demonstrated that Ge-132 could obviously enhance growth of CFU-GM in vitro when 0.05μg/ml of Ge-132 was added. As the dosages of Ge-132 increased, the number of CFU-GM increased still more, up to the dosage of 500μg/ml. The best stimulating dosage was 5μg/ml. Depletion of adherent cells reduced the effect of Ge-132 on colony growth. Depletion of adherent cells and T lymphocytes at the same time abolished the effect. Our studies indicate that the stimulating effect of Ge-132 on human marrow CFU-GM may be mediated indirectly by the interaction of T lymphocytes and monocytes.
