酶联免疫印迹对弓形虫抗原组分的研究

摘要 用酶联免疫印迹(ELIB)对弓形虫抗原纽分分析的结果显示:经SDS-PAGE后的弓形虫抗原组分分别为17.8KD、21.3KD、30KD、36.5KD、39.1KD和50.2KD。电转印至硝酸纤维膜上的各抗原组分,用ELIB分析,弓形虫感染宿主的血清能与抗原组分蛋白起反应,其中39.1KD和21.3KD分子量组分蛋白带显色最深、提示这二种组分蛋白是弓形虫IgG抗体识别的主要组分蛋白,而对照血清与各组分蛋白均为阴性。本文研究证实ELIB检测弓形虫感染宿主,其敏感性和特异牲均较高。 Antigenic components of toxoplasma were determined by enzyme-linked immunoblotting technique(ELIB).The results showed that antigen protein bands were distributed under 50.2KD fields and had six bands that molecular weight were 17.8KD,21.3KD,30.0KD,36.5KD,39.1KD and 50.2KD respectively.The array of resolved antigen protein were electrophoretically transferred nitrocellulose filter paper and incubated with serum sample from rabbits with Toxoplasma gondii infections or with serum sample from seronegative controls,Serum immunoglobulin G bound to individual toxoplasma antigen were detected by HRP-stsphylococcal protein A.A complex array of toxoplasma antigens was revealed by this procedure.Immunoglobulin G directed against a low-molecular-weight antigen(two major antigens with apparent m.w.of 39100 and 21300)was detected in sera from rabbits infected with toxoplasma(10 sera tested)and absent from sera from individuals with seronegative controls(10 healthy rabbits,10 healthy persons,8 patients with entamoeba abscess of liver,10 rabbits infected with Schistosoma japonicum,3 patients with Plasmodium vivax infection).
